The overall goal of this Core laboratory is to provide the infrastructure, materials, animals, technical expertise and support that will facilitate the use of humanized immunodeficient mice in studies examining the pathogenesis and treatment of HIV-induced disease. Recent advances have enabled the recapitulation of human immune cell development and function in immunodeficient mice. Human hematopoietic tissue is placed in the mice where it undergoes multilineage hematopoiesis into multiple types of human immune cells. The human tissue utilized in these studies is capable of a high degree of experimental manipulation prior to and subsequent to implantation in the mouse and is further susceptible to infection with HIV. This provides a powerful surrogate in vivo model to examine human hematopoiesis, the pathogenic effects of HIV infection, and ways to protect the human immune system from HIV.
The generation of these humanized mice is a highly specialized procedure, due to the requirement for immunodeficient mouse strains, human hematopoietic tissue, infectious material, specialized facilities, and the necessary skill and knowledge to perform experiments in this system. In addition, due to the lower breeding rates of immunodeficient animals and the surgical manipulation of human tissue, the costs of performing experiments in these systems are significantly higher than costs associated with non-immunodeficient experimental mouse work. The central purpose of the UCLA CFAR Humanized Mouse Core Laboratory is to provide the environment, technical expertise, and resources to facilitate AIDS- and cancer- related humanized mouse based studies and development.

The core can also provide consultation in flow cytometry, immuno-histochemistry, PCR and other assays. However, final analysis of experimental animals is the responsibility of the individual investigator. Some of these techniques are available through other UCLA AIDS Institute/CFAR core facilities.
